It can be done without the audience detecting the difference. So, yes, to some extent the talking voice is different than the singing voice. To comment more directly on what I think your question may be... I think that if a single puppeteer does all the voices in a production, their various singing voices tends to merge towards a single "voice". I think a lot of the individual's character is maintained by way the song is written, and less so by the way he sings an "Aaaaaaah". Still, there is room for a singing voice to be more nasal, soft, or harsher than other characters.
